2. An Attempt to Improve Bovine Mycoplasmatales Primary Isolation.
The sensitivity to 30 antibiotics of Mycoplasma (10 strains) and Acholeplasma (8 strains) was tested in an attempt to find a better one for suppression of contamination in the culture media, used for primary isolation of Mycoplasmatales. The most effective antibiotics for suppression of Gram-positive and Gram-negative organisms and did not inhibit Mycoplasma

3. Detection of multiple mycoplasma infection in cell cultures by PCR
A total of 301 cell cultures from 15 laboratories were monitored for mycoplasma (Mollicutes) using PCR and culture methodology. The infection was detected in the cell culture collection of 12 laboratories. PCR for Mollicutes detected these bacteria in 93 (30.9%) samples. Although the infection was confirmed by culture for 69 (22.9%) samples, PCR with generic

4. Avaliação clínica e estudo da ocorrência de fêmeas ovinas infectadas por micoplasmas na região de Piedade - SP - "Avaliação da infecção sobre a produtividade do rebanho" / Clinical avaliation and ocurrence studies of mycoplasmas infected ovine female in the Piedade region - State of São Paulo- "Avaliation of the infection in the productivity of the herd"
Sheep industry is growing rapidly in Brazil. However many diseases are the principal bottleneck to the increasing these activities. Among the reproductive diseases the vulvo-vaginites caused by Mycoplasma play an important role. Although no studies has been carried out in brazilian sheep herd so far. For that 60 ewes were ramdomly from different flocks selec

5. Respiration-associated components of Mollicutes.
No cytochrome pigments were detected by difference (reduced minus oxidized) spectroscopy at liquid nitrogen temperature in whole-cell preparations or membrane fractions of Acholeplasma axanthum S273, Acholeplasma equifetale N93, Acholeplasma granularum BTS39, Acholeplasma laidlawii B-PG9, Acholeplasma modicum PG-49, Acholeplasma oculi 19L, Mycoplasma arginin

6. DNases of Acholeplasma spp.
One strain from each of seven species of Acholeplasma was examined for the presence of DNases. Six of the strains were found to be DNase positive when assayed with DNA-methyl-green-containing agar, indicating that this method can be used to differentiate the seventh strain, Acholeplasma axanthum, from the remaining Acholeplasma spp. Electrophoretic patterns

7. Identification of the major antigenic determinants on lipoglycans from Acholeplasma granularum and Acholeplasma axanthum.
The major antigenic determinants of the lipoglycans from Acholeplasma granularum and Acholeplasma axanthum were found to be the oligosaccharide sequences Glcp(beta 1 leads to 2)-Glcp(alpha 1 leads to 4)-Glcp and Glcp(beta 1 leads to 2)-Glcp(beta 1 leads to 2)-Glcp, respectively. The disaccharides sophorose and maltose inhibited hemagglutination by specific a

8. Nisin resistance distinguishes Mycoplasma spp. from Acholeplasma spp. and provides a basis for selective growth media.
The sensitivity of 11 Mycoplasma and 5 Acholeplasma species to the bacteriocin nisin was determined. When applied on filter paper discs to lawns of acholeplasma cells, nisin (20 nmol per disc) gave 3.5- to 7.0-mm zones of growth inhibition. The inclusion of 0.2 mM nisin in agar medium reduced the number of Acholeplasma laidlawii colonies by a factor of more

9. Acholeplasma axanthum, sp. n.: a New Sterol-Nonrequiring Member of the Mycoplasmatales
Mycoplasmas recovered from tissue cultures and previously shown to belong to the sterol-nonrequiring group of mycoplasmas have been further characterized. The biological and serological properties of these strains show them to be clearly distinct from Acholeplasma laidlawii and A. granularum, two species of sterol-nonrequiring mycoplasmas recently reclassifi

10. Nucleic acid relationships among Acholeplasma species.
3H-labeled Acholeplasma DNA probes were generated in vitro by the nick-translation method and used to determine the nucleotide sequence homology among the type strains of the eight currently recognized species of Acholeplasma. Very little nucleotide sequence homology (less than or equal to 18%) was found among the eight species, with heteroduplexes showing a

11. Presence of protein constituents of the gram-positive bacterial phosphotransferase regulatory system in Acholeplasma laidlawii.
Acholeplasma species have been reported to lack a functional phosphoenolpyruvate:sugar phosphotransferase system (PTS). We show here that Acholeplasma laidlawii possesses activities of enzyme I, HPr, HPr(ser) kinase, and HPr(ser-P) phosphatase but lacks detectable activities of enzymes II of the PTS. HPr from this organism was purified, and the regulatory pr

12. Interaction of mycoplasma virus type 2 with cellular components of Acholeplasma laidlawii strain JA1.
Mycoplasma virus type 2 was shown to adsorb specifically to intact cells, membranes, and lipoglycan of Acholeplasma laidlawii strain JA1 but not to these components of Acholeplasma oculi. The oligosaccharide chain of the lipoglycan defined the specificity of the receptor site since deacylation not only did not reduce adsorption but increased it threefold. Ac
