The increasing demand for production of pharmaceutical interest recombinant proteins has been the goal for the development of additional systems to its production on a large scale and reduced costs. Eukaryotes expression systems are possible alternatives to the production of recombinant protein (Leite, 2000). The secretion of these polypeptides in the milk of transgenic animals promised high level of heterologous pharmaceutical products as well as the correct process and post-translation modifications (Kerr et al., 1996). The production on eukaryotic systems has presented characteristic interesting in such a way with regard to the maintenance of its process of synthesis, envelopment and processing, and to the significant reduction of costs, increase of stability, maintenance of biological activity, absence of contaminants and pathogens to the humans (Kusnadi et al., 1997). Blood coagulation factor IX is a serine-protease glycoprotein and it presents molecular mass of 55kDa. The polypeptide is synthesized in the liver and secreted in the blood, where it participates of reactions group involving coagulation factors and proteins that permit the blood coagulation (Thompson et al., 1986). Genetic alterations that cause the dysfunction or the absence of synthesis of the FIX lead to the hemofilia B, a recessive illness linked to chromosome X which affects about 1 in each 10,000 men in Brazil. In this work two lineages of mice (Mus muscullus) were microinjected with the human coagulation factor IX gene. The founding females and its progenies secreted the recombinant FIX in their milk (3% Total Soluble Protein). The stable integration of transgene - under control of the milk -casein pBC1 promoter - was confirmed by southern blot analysis. The presence of the FIX recombinant protein in the milk of transgenic females was verified by western blot. Tests of its activity in B hemophilic blood samples were confirmed by coagulometer pattern, validating this heterologous system.
