AvaliaÃÃo de aberraÃÃes cromossÃmicas em bovinos (Bos taurus taurus) infectados pelo papilomavÃrus bovino

RESUMO
Bovine papillomavirus (BPV) is represented by 10 papillomavirus subtypes, epiteliotropic or mucosotropic, which are transmitted among bovines through contaminated blood and ephitelial contact. This virus may be in a latent form on host lymphocytes, triggering chromosomal instabilities. In this context, this study aimed to evaluate chromosomal aberrations which might result from this process of instability. Peripheral blood samples were collected from Bos Taurus females.Viral detection was performed using polymerase chain reaction technique (PCR) and lymphocytes cultures were established for cytogenetic analysis. 61 animals were analyzed in âblind-testâ in slides submitted to conventional staining and C-banding. Twenty eight (28) animals were verified as no infected by BPV. Thirty three (33) animals presented BPV type 1 and /or 2. The BPV infected group was subdivided into two subgroups: symptomatic and asymptomatic. Symptomatic group (with papillomatosis) represented by 17 females, exhibited an average of 42.71% aberrant cells per individual.Asymptomatic group (without papillomatosis), represented by 16 animals exhibited an average of 40.19%. The analysis included 50 to 100 cells per sample, totalizing 2203 cells: 918 presented one or more chromosomal aberrations. The chromosomal aberration rate in symptomatic and in asymptomatic animals was respectively 42.7Â 7.8 and 40.2 Â 11, compared with aberration rate of control group,4Â2. To analyze statistical differences among these three groups, were used Kruskal-Wallisâ Test followed by Mann-Whitneyâs Test. These analysis reported that aberrant cells variability between the control group and BPVinfected groups was larger on infected cattle group (P <0,0001). However, significant differences were not observed between infected subgroups (P = 0,62). The identified chromosomal aberration types were: centric association (CA); acentric fragment (AF); telomeric association (TA); telomeric association by a unique chromatid (TAcr); chromatid breaks (CtB); chromossomic breaks (CmB); gaps; aneuploidy; polyploidy; addition or loss of chromosomal segment (add or del) and early chromatid separation (EcrS). The obtained data demonstrated that the high rates of cromossomal aberrations (numeric and structural) are related to BPV action, allowing distinction between infected and not infected animals.
