RESUMO

Osteoporosis is a progressive systemic skeletal disease, characterized by bone mass reduction and deterioration of the bone micro-architecture, leading to fragility and with consequent increase of fracture susceptibility. The preliminary and secondary target of pharmacotherapy in osteoporosis is to prevent bone fractures. Initial treatment of osteoporosis must be done through calcium supplementation. The most recommended is calcium carbonate, because it has greater amount of elementary calcium (40%). Oyster shell powder is a feasible and effective alternative in this mineralâs supplementation, due to being mostly made up of calcium carbonate. It is of utmost importance to evaluate all the characteristics of powder and granulates, among them, mean size of the particles, particle size distribution, particle shape, surface structure, surface area, among others, to determine the parameters involved in the development process of the tabletâs dosage form. Any factor that changes the desegregation and dissolution processes can directly affect the bioavailability, expressed in terms of quantity of drug absorbed and speed of the absorption process. A way of evaluating the in vitro availability of this mineral is through the dissolution test, which is sensitive to some changes occurred during the obtainment process of the tablets. In light of this, this work was aimed at evaluating the parameters that could interfere in the drugâs dissolution. A factorial planning was conducted for this, obtaining different granulates and tablets with different characteristics, characterizing them physically and observing the influence of each parameter modified in the dissolution of the tablets. Furthermore, an analytical method was developed and validated for dosing of the calcium carbonate in the raw material, finished product and to quantify the dissolved calcium carbonate content. The methods were sturdy, exact and precise, with reproducible and reliable results
