Disfunção na resposta ao estresse do retículo endoplasmático em linfócitos de pacientes com transtorno de humor bipolar

RESUMO

Bipolar disorder (BD) is a severe chronic psychiatric disorder that has been related to several cellular dysfunctions, such as an impaired endoplasmic reticulum (ER) stress response. We aimed to evaluate the unfolded protein response (UPR) and the outcome of this cellular process in cultured lymphocytes from patients with BD and healthy subjects, assessing its relationship to the stage of the disorder. Lymphocytes from 30 BD patients and 32 age- and sex-matched controls were treated with tunicamycin, an ER stressor, for 12h or 24h in order to measure levels of UPR-related proteins (GRP78, eIF2¿-P and CHOP) by flow cytometry, and for 48h to analyze ER stress-induced cell death. The effect of lithium on these parameters in cultured lymphocytes was also evaluated. In the control group, inductions of GRP78 and eIF2¿-P by tunicamycin after 12h and 24h and of CHOP after 24h were observed. Tunicamycin-induced increases in UPR-related proteins were not found in the BD group. Among euthymic patients, those at an early stage of disorder had a better response to ER stress when compared to patients in advanced stages, with GRP78 and eIF2¿-P levels similar to controls after 12h of treatment with tunicamycin. Tunicamycin-induced cell death was higher in BD patients compared to controls. Lithium did not induce differences in the expression of the evaluated proteins and did not reverse tunicamicin-induced cell death, but led to a small yet statistically significant decrease in this parameter. This study extends earlier findings about impaired ER stress response in primary cultures of lymphocytes from BD patients. ER dysfunction may be associated with decreased cellular resilience in BD, ultimately contributing to the progression of the disorder.
