Influencia da evolução do tecido granulomatoso sobre a biodisponibilidade da amoxicilina : estudo ex vivo, em ratos

Since the penicillin was discovered many studies have been carried out in order to justify its large use, however in some aspects about pharmacokinetics properties have not yet well understood. The subject of this study was to evaluate the penetration of amoxicillin in chronic lesion, as well as in serum levels. We used 160 rats with four PVC sponges previously implanted in the back. After 7, 14, 21 and 28 days of sponge introduction, the animais received 3.57 mg/kg, 7.14 mg/kg, 40 mg/kg and 80 mg/kg of p.o. amoxicillin. Control animais received NaCI 0.9% solution. One hour often the administration of drug, the tissue and blood samples were collected and were placed on Petri dishes, previously inoculated with 108 c.f.U. of Staphy/ococcus aureus (A TCC 25923) in Mueller Hinton agar. The blood samples were centrifuged and the serum (10 j.JL) was placed on 6.25 mm (diameter) paper disks. After 18 hours of incubation the diameters of inhibition zones were measured and registered for later analysis. A regression curve had been was made with the diameters of inhibition zones and the log of concentration of drug. The variance analysis showed no statistically significant differences (a=0,01) between the time periods and the ~oses of amoxicillin. The results suggest that the penetration of the drug is not changed by the time of development of the tissue
