Influencia de antiinflamatorios sobre a concentração serica e tecidual da amoxicilina : estudo ex-vivo, em ratos

RESUMO

The prescription of antibiotics associated to anti-inflammatory is common in dentistry and its effects have been studied. Thus, the aim of this work was to evaluate the interaction among amoxicillin, sodium diclofenac and betametasone in tissues and serum, utilizing ex-vívo model. It was used 56 male Wistar rats (7 groups, n= 8), six weeks old, kept in acclimatized environment with food and water ad líbítum. It was implanted four sponges of PVC under the back skin of each animal. Seven days later, it was administered p.o.: amoxicillin 25mg/kg (group 1), sodium diclofenac 2.5mg/kg (group 2), betametasone 0.1mg/Kg (group 3), sodium diclofenac 2.5 mg/Kg + amoxicillin 25mg/Kg (group 4), betametasone 0.1mg/kg + amoxicillin 25mg/Kg (group 5), sodium diclofenac 2.5mg/Kg + betametasone 0.1mg/Kg (group 6) and 1.0mL of solution of sodium chloride 0.9% (group 7). After 90 minutes, the granulomatous tissues were collected and placed in MHA with S. aureus 108 c.f.u/mL. At the same time the blood was collected, centrifuged and 30µL of the serum placed in MHA with S. aureus 108 c.f.u/mL After 18 hours of incubation, the inhibition zones were measured. The tissue inhibition zones of the interaction between amoxicillin and sodium diclofenac showed a medium value of 21.44mm and a tissue concentration of 0.42µg/g, while amoxicillin and betametasone association showed 26.69mm and a tissue concentration of 1.39µg/g. The serum inhibition zones of the interaction between amoxicillin and sodium diclofenac showed a medium value of 21.19mm and a serum concentration of O.42µg/mL, while amoxicillin and betametasone association showed 25.53mm and a serum concentration of 1.41µg/mL. The amoxicillin and sodium diclofenac association had presented a lower tissue and serum concentration of the antibiotic in relation to its association with betametasone
