Perfil eletroforetico de proteinas intracelulares de Candida albicans isoladas da cavidade bucal e outros sitios anatomicos de humanos

RESUMO

Samples of Candida albicans isolated from the human oral cavities and other anatomic sites, were studied. The purpose of the present research was to study the degree of similarity among these isolates through the whole-cell polyacrylamide gel electrophoresis technique (SDS-PAGE). The samples were inoculated in YPD culture medium, and after the incubation, they were centrifuged at 3,000 x g for 5 minutes, with the sediments were washed in cold distilled water and whole-cell proteins were extracted in liquid nitrogen. The cell extracts were submitted to SDS-PAGE technique and after the electrophoresis, the gels were revealed and the protein bands were submitted to computacional analysis on the NTSYS-pc 1.70 version statistical package applying the coefficient similarity SM (Simple Matching Coefficient). Those similarity matrices were treated by the UPGMA algorithm, allowing the analyses of similarity levels among C. albicans isolates by dendrograms. The eiectrophoretic analysis of the C. albicans proteins extracts isolated from oral cavities and others human anatomies sites, allowed the construction of dendrograms including clustering of isolates in representatives groups of species. Samples of C. albicans species of the same anatomic site showed similarity values up 80% (SSm = 0,800) supporting the species grouping. Whole sample of these organisms (all anatomic sites) selected randomly including the type-strain CBS-562 showed high similarity. The clusters composition expressed by the dendrograms of all the sample were not dependent of factor as: anatomic sites and studied persons
