Peritrophic membrane origin ultramorfology and ultra structural analysis in bees (Hymenoptera) / Ultra-morfologia e análise ultraestrutural sobre a origem da membrana peritrófica em abelhas (Hymenoptera)

RESUMO

Two experiments were carried out about bees peritophic membrane (PM). This not cellule structure, quitine fibril composed, glicosaminoglicanes and protein, to cover midgut inner. The first experiment was carried out through scanning microscopy and had to objective describe ecto and endoperitrophic spaces characteristics in solitary and eusocial bees. Adults bees midguts were utilized, it were prepared, dehydrated in a graded ethanol series (70, 80, 90, 95 e 99,9%) and transferred to hexamethyldisilazane, air dried and analyzed in LEO VP1430 SEM. Results showed in Mliípona quadrifasciata anthidioides (Meliponini) the PM has two layers, whereas ectoperitrophic space, turned midgut, showed hexagonal ornamentation and the surface associated with the endoperitrophic space is folded. Nannotrigona sp. (Meliponini) PM the endoperitrophic space showed overload irregular fragment. In Apis mellifera (Apini), Centris (Melanocentris) sp. (Centridini) and Epicharis flava (Centridini) the PM showed lamellar aspect. In Megachile sp. (Megachilidae) and Oxaea flavescens (Andrenidae) the PM showed irregular layers with dense material covering their surface. The second experiment was carried out with solitary and eusocial adults bees midgut. The objective this work was to identified the PM origin, well this structure in many insects species can midgut or cardia cellules synthesized. The localization of the PM production sites was realized using antibody produced against peritrophin, PM exclusive protein. The results showed peritrophin production sites in every midgut cellule, including cardian cellule of the Meliponini tribe bees. Therefore, analyzed species Auglochlora sp (Halictidae), Megachile sp. (Megachilidae), Oxaea flavescens (Andrenidae), Apis mellifera (Apidae: Apini), Bombus morio (Apidae: Bombini), Centris (Melanocentris) sp. (Apidae: Centridini) and Epicharis flava (Apidae: Centridini) the PM is produced only to midgut digestive cellules, type I classified. In Melipona quadrifasciata anthidioides (Apidae: Meliponini), Frieseomelita varia (Apidae: Meliponini) the PM two origin presented, as many midgut digestive cellules, type I PM, as cardian region cellule, type II PM.
